中文摘要: 细胞周期调节异常和凋亡是肿瘤的重要特征,也是肿瘤研究的一个重要方向。APMCF1是我们在凋亡的MCF-7细胞cDNA文库中克隆的一个全新基因,该基因在凋亡的MCF-7细胞中表达明显上调,另外,其在肝癌细胞系HHCC和HepG2中分别过表达和Knockdown后均显著影响细胞周期,提示它很可能通过调控细胞周期从而影响细胞凋亡,我们进一步发现并证实APMCF1和SGTA存在相互作用,而文献报道也表明SGTA与细胞凋亡、增殖分化、细胞周期阻滞密切相关,因此进一步深入研究成果值得期待。蛋白质相互作用研究是研究基因功能的一个有效途径,据此我们拟进一步明确APMCF1与SGTA相互作用位点、上下游关系及其相互作用在肝癌细胞周期调节及凋亡中的作用机制,以深入了解APMCF1和SGTA的功能,结果也将有助于深入了解肝癌的发生机制。

英文摘要: The anomaly of cycle regulation and apoptosis of cells are the main characteristics of tumours as well as important fields of research in tumour studies. APMCF1 was a new gene we cloned in the cDNA library of cell MCF-7 and was found to be significantly upregulated in the expression of the apoptosis of cell MCF-7 in the experiment. In addition, the overexpression and knockdown of expression greatly influenced the cell cycles of HHCC and HepG2 cell lines, respectively, so APMCF1 was very probably a gene related to the cycle regulation or to the apoptosis of cells and deserved to be studied further. It was also found that there was an interaction between APMCF1 and SGTA, and earlier studies also found that SGTA was closely related to cell apoptosis, cell proliferation and differentiation, cell cylce arrest. The research into the interaction between proteins was effective in the study of gene functions, based on which we could find APMCF1 and SGTA interaction sites, the relationship between upstream and downstream as well as the mechanism of their interaction in hepatocarcinoma apoptosis and cell cycle regulation in order to learn about the functions of APMCF1 and SGTA. The findings will also help to learn more about the the occurrence mechanisms of hepatocarcinoma.
