中文摘要: 失眠严重影响人们的生活质量,中药中含有大量的镇静安神、促睡眠活性成分,但这些中药如何发挥镇静安神、促睡眠作用并不很清楚。 以Melatonin 受体、Serotonin受体、Orexin受体、Histamine受体、Adenosine 受体为代表的五类G蛋白偶联受体调控着人体的睡眠。本研究拟构建可感知上述五类受体空间构象变化的G蛋白偶联受体分子探针,以这些受体分子探针为平台,通过监测受体分子内荧光共振能量转移基线的波动,对有镇静安神、促睡眠作用的中药进行全方位的筛选,并从受体激活动力学水平、钙离子释放水平和ERK激活水平阐释这些活性成分的分子作用机理。本项目将能筛选到多种具有多重功效的受体激动剂、拮抗剂或反向激动剂,为研发有完全自主知识产权的促睡眠新药奠定基础。更为重要的是,本研究将为中药活性成分的筛选创建新的方法学平台,为阐释中药的作用机理开辟新的思路。

英文摘要: Insomnia plays a negative role in the quality of modern life. Chinese medicines contain vast amount of tranquilizing, sleep promoting components. But it is not so clear how these Chinese medicines tranquilize brain and promote sleep. Five G protein-coupled receptor (GPCR) families (Melatonin receptor family, Serotonin receptor family, Orexin receptor family, Histamine receptor family and Adenosine receptor family) regulate sleep. In this study, we will construct five groups of Intramolecular GPCR fluorescence resonance energy transfer (FRET) sensors, which can detect the spatial change of receptors. Using these sensors as drug screen platform, and the change of FRET baseline as an indicator, we will carry out comprehensive screening of tranquilizing, sleep promoting components from Chinese medicines. Our work will find multifunctional receptor agonists, antagonists or inverse agonists, in the same time, our work will elucidate the tranquilizing mechanism of these active components by examining receptor kinetics, cell calcium level and ERK activity. More importantly, our work will create a new drug screen platform and give new scientific insight into the function of Chinese medicines.
