中文摘要: 经颅超声已被证实可以促进脑损伤后的神经可塑性变化,但确切的分子调控机制尚不明确。我们应用miRNA芯片筛选到经颅超声治疗后miR-132在脑瘫大鼠海马中表达显著上调,并通过Real-time PCR验证证实。目前已知miR-132参与p250GAP、NR2B等突触可塑性相关基因调控,因此我们提出经颅超声通过调控miR-132促进海马突触可塑性实现功能康复的假设。本研究拟利用行为学、病理学、分子生物学等技术进行体内外研究:观察经颅超声治疗后海马突触可塑性变化及miR-132、下游靶基因表达变化;进一步评价miR-132过表达/表达沉默对超声相关作用的影响,以期证实经颅超声促进神经可塑性变化的机制之一是通过调控miR-132促海马突触可塑性途径来实现的。经颅超声的miRNA机制未见报道,课题将从全新视角提出经颅超声康复治疗的理论依据。

英文摘要: Transcranial ultrasound has been shown to promote neural plasticity after brain injury, but the exact molecular mechanism remains unclear. Using miRNA microarray analysis, we found miR-132 expression was significantly upregulated in the hippocampus after ultrasound therapy in rats with cerebral palsy, and the resulting data confirmed in the real-time PCR experiments. MiR-132 is known to participate the regulation of synaptic plasticity-related genes, such as p250GAP and NR2B, so our hypothesis propose that the transcranial ultrasound promote hippocampal synaptic plasticity and functional recovery via miR-132. In this study, we use behavioral, pathological and molecular biological techniques in vivo and in vitro to develop scientific research. First, we observe the hippocampal synaptic plasticity, the expression of miR-132 and its downstream targets changes after transcranial ultrasound therapy. Second, with the miR-132 gene overexpression or silencing, the relationship between miR-132 and synaptic plasticity of ultrasound will be investigated. These result are benefit for us to clarify whether one of the mechanisms of transcranial ultrasound in promoting neural plasticity contribute to miR-132. The role of miRNA on transcranial ultrasound has not been reported, so this study will present new theoretical perspectives for transcranial ultrasound on facilitating rehabilitation.
