中文摘要: 足细胞损伤是糖尿病肾病发生发展的基础,目前研究证实足细胞通过自噬维持自身稳态。而我们前期证实,自噬可以通过能量代谢相关通路(如:mTOR信号通路)调控嘌呤霉素引起的足细胞损伤,但自噬是否可以介导高糖环境下的足细胞损伤,从而影响糖尿病肾病的发生,尚不清楚。我们推测,自噬可以通过调控与能量代谢相关的信号通路,介导糖尿病肾病时足细胞的损伤。本研究通过体内及体外研究,采用基因转染与RNA干扰技术,阐明高糖致肾小球足细胞损伤的过程中,自噬与能量代谢相关通路之间的关系,明确参与肾小球足细胞自噬的信号通路。且在体外及体内,通过阻断RAS系统,明确高糖与肾小球内及足细胞RAS系统的激活在自噬诱导足细胞损伤中的协同作用。并进一步明确针对足细胞自噬调控的药物可以成为预防及治疗糖尿病肾病的新靶点的可能性。研究成果将深入解析自噬调控肾小球足细胞损伤的分子机制,为阻止糖尿病肾病的发生发展提供新的干预靶标。

英文摘要: Podocyte injury is the basis of occurrence and development of diabetic nephropathy, while autophagy maintains podocyte homeostasis. Our previous studies have confirmed that autophagy could regulate puromycin-induced podocyte injury by energy metabolism related signaling pathway (eg. mTOR signal pathway). However, we do not know whether autophagy can regulate high glucose-induced podocyte injury, thus would lead diabetic nephropathy. We hypothesized that, autophagy mediated podocyte injury in diabetic nephropathy by regulating of energy metabolism related signaling pathways. This study will elucidate the relationships between autophagy and energy metabolism related signaling pathway in high glucose-induced glomerular podocyte injury using gene transfection with RNA interference technology in vivo and in vitro. Through these, we will clarify the important signaling pathway in podocytes autophagy. And we will make clear the synergistic effect in RAS system activation and autophagy induced podocyte injury in high glucose by blocking RAS system in vivo and in vitro, respectively. And then, we will make sure the possibility of drugs for regulation in podocyte autophagy could prevent and treat diabetic nephropathy. The research results will in-depth analyze the molecular mechanisms in autophagy mediated podocyte injury
