中文摘要: 生理状态下卵巢分泌的雌激素反馈调节卵泡闭锁或成熟,而卵泡闭锁最重要的诱因是细胞凋亡。不同组织中雌激素与其不同受体(ERα/ERβ)结合,促进或抑制细胞凋亡。申请人前期研究发现,雌激素能上调大鼠脑中雌激素受体和Bcl-2的表达,抑制神经元凋亡;近期研究结果表明,牦牛子宫中有ERα的表达,并随发情周期不同出现规律性变化。但有关牦牛卵巢中雌激素受体的表达及卵泡发育过程中细胞凋亡的机制未见相关报道。因此,本项目拟以牦牛为研究对象,利用免疫组织化学、原位杂交和RT-PCR等方法,通过在体和离体研究,探讨雌激素对卵巢中雌激素受体(ERα/ERβ)表达的调控机制及其与卵巢细胞凋亡和凋亡相关基因(Bcl-2/Bax,Fas/FasL,Caspase-1,Caspase-3,Caspase-9)表达的相互关系,阐明雌激素对牦牛卵泡发育过程中细胞凋亡的调控机制,为提高牦牛卵巢储备能力和繁殖性能提供新思路。

英文摘要: Estrogen secreted by ovarian feedback-regulates follicular atresia or maturation, and follicular atresia caused mostly by cell apoptosis. In different tissue, estrogen binding to its different receptor subtypes(ERα/ERβ)could promote or inhibit cell apoptosis.Our Previous studies showed respectively that, in rat brain estrogen could up-regulate the expression of ER, and could up-regulate Bcl-2;expression of ERα in yak uterus were differed with stages of the estrous cycle.But,there is no any report about expression of ER in yak ovay, and the mechenism of cell apoptosis during yak follicular development is still unclear.In order to investigate the mechanism of estrogen regulating cell apoptosis during yak follicular maturation, and lay a theoretical foundation to improve yak ovarian reserve capacity and yak fecundity, in this project, effect of estrogen on expression of ER subtypes, apoptosis relative genes (including Bcl-2/Bax,Fas/FasL,Caspase-1,Caspase-3,Caspase-9) and cell apoptosis in yak ovay during follicular development will be analyzed by immunohistochemistry, hybridization in situ and RT-PCR in vivo and in vitro.
