中文摘要: 血清中多种肿瘤标志物的同时检测对于降低癌症误诊几率、提高诊断准确性有着十分重要的意义。利用荧光纳米簇光稳定性强的优点,结合具有特异性识别作用的适配体,能够构建灵敏度极高的 off-on型荧光共振能量转移(FRET)适配体传感器。基于荧光纳米簇作为能量供体的off-on”型FRET适配体传感器在肿瘤标志物检测方面具有很大优势,但目前利用此类传感器对多种肿瘤标志物进行同时检测尚未见报道。本研究拟以不同的蛋白质、核酸分子、谷胱甘肽等为模板,合成Au、Ag、C、Cu等不同的荧光纳米簇,通过控制反应条件以获得不同发射波长的荧光纳米簇,并在其表面修饰针对不同肿瘤具有特定识别的适配体,筛选出合适的几组功能化荧光纳米簇构建off-on”型FRET的适配体传感器,实现血清中几种肿瘤标志物的同时检测。本方法快速、准确且灵敏度高,能够同时检测血清中多种肿瘤标志物,将为癌症的准确诊断提供有力的技术支持。

英文摘要: It is of great importance to simultaneously detect multiple tumor biomarkers in serum in order to improve the accuracy of cancer diagnosis. Owing to their excellent light stability and high sensitivity, “off-on” aptamer sensors constructed with fluorescent nanoclusters and aptamer has obvious advantages for the detection of tumor biomarkers based on the fluorescence resonance energy transfer (FRET). However, there has no report on the application of fluorescent nanoclusters-aptamer sensor for the simultaneous detection of several tumor biomarkers. In this proposed research, we will synthesize a series of fluorescent nanoclusters (Au, Ag, C, Cu) with different emission wavelengths protected by proteins, DNA and GSH, which will be further modified with specific aptamers. We will then select several appropriate functionalized fluorescent nanoclusters to construct the “off-on” fluorescent nanoclusters-aptamer FRET sensors for simultaneous detection of different tumor biomarkers in human serum. The proposed method can be used for the simultaneous detection of several tumor biomarkers in human serum with high sensitivity and accuracy, and it may become a powerful tool for cancer diagnosis.
