中文摘要: 表观遗传修饰是的控制大脑细胞内基因开和关的重要化学过程,可以部分解释正常大脑细胞分工的不同,也有助于更好地理解某些精神疾病的过程。本项目在亚急性、慢性抑郁症动物模型上,探讨针刺的调整效应可能发生在基因表达调控的上游- - 表观遗传修饰层面,从而进一步调整抑郁症相关脑区的病理改变,从而发挥整体调节作用。已取得下列研究结果: 1)建立了亚急性、慢性抑郁症大鼠模型。 2)同时,在慢性抑郁大鼠模型中,长期给予电针治疗,观察到电针的抗抑郁效应。 3)检测抑郁模型大鼠海马区表观调控变化,观察到这些表观调控变化可能与下游的蛋白表达调控相关。 4)同时观察到抑郁模型大鼠海马星胶大量丢失,胶质源性神经营养因子减少,而给予电针治疗后可以显著改善抑郁模型大鼠海马内这一显著的病理性改变。 以上工作进一步加深了对抑郁症和针刺抗抑郁机理的理解,也为临床上抑郁症的中西医结合综合治疗提供新思路。

英文摘要: Epigenetics controls the on-off of the gene expression and affects normal brain processes - such as development or memory. And epigenetic changes are also a part of brain diseases such as mental illness and addiction. Understanding the role of epigenetics in brain disease may open the door to being able to influence it. This may lead to the development of new and more effective treatments for brain diseases. This project was focus on the epigenetical modification and its role in the antidepressant effect of electroacupuncture. The results were exhibited as: 1) Established the sub-acute and chronic rat model of depression 2) Observed the antidepressant effect of EA in the rat model of depression 3) Examined the epigenetical modification in rat model of depression through the high-throughput DNA methylation microarray and the down-stream mRNA and protein expression through real-time PCR and western blot. 4) Further, observed the glia atrophy and the down-regulation of GDNF in depression model and EA attenuated the pathophysiologic changes. These studies are helpful for furthering investigation of the mechanisms underlying depression and the antidepressant effect of acupuncture.
