中文摘要: 幽门螺杆菌(Hp)感染严重危害着人类的健康安全,Hp疫苗的研制对于Hp感染的预防将起重要作用。课题组在大肠杆菌表达体系成功克隆、表达了多个幽门螺杆菌疫苗候选分子(尿素酶B,UreB;中性粒细胞激活蛋白,NAP;分泌蛋白,SP),并将UreB、NAP成功在枯草芽孢上表达,使用表面表达Ure枯草芽孢进行了小鼠的攻击感染实验。结果显示,表面表达Ure的重组芽孢腹腔注射免疫小鼠可产生特异性体液免疫,两周后血清特异性IgG显著高于非重组芽孢组及空白对照组P<0.05;使用表面表达Ure的重组芽孢口服免疫小鼠血清特异性IgG也显著高于对照组表P<0.05,表明重组芽孢具免疫原性,注射及口服均可引起小鼠系统免疫;口服重组芽孢组小鼠粪便特异性IgA水平显著高于对照组P<0.05。并且,口服UreB重组芽孢对Hp攻击具一定的保护作用,免疫保护率为62.5%(5/8)。

英文摘要: Helicobacter pylori ( Hp ) infection is harmful to human health and safety, Hp vaccine research will play an important role for prevention of Hp infection.multi Helicobacter pylori vaccine candidate molecules (urease B,UreB;neutrophil activating protein NAP, secreted protein, SP )were cloned, expressed in Escherichia coli expression system successfully.UreB,NAP were surface expressed in Bacillus subtilis spore and recombinant UreB spores were used to immunize mice.The animal experiment results showed: mice immunized intraperitoneally with surface expression of Ure recombinant spores can produce specific humoral immunity, serum specific IgG is significantly higher than that of control group (P < 0.05) two weeks after immunization, serum specific IgG in oral immunization with recombinant Ure spores mice was significantly higher than that in control group, P < 0.05, indicating that recombination bacillus was immunogenicity,injection intraperitoneally and oral administration can induce immune reaction; also fecal specific IgA level in oral administration of recombinant spore mice group was significantly higher than those in control group, P < 0.05. And, oral administration of UreB recombinant spore can produce protective effect against Hp challenge, the immune protective rate was 62.5% (5/8)in mice.
