中文摘要: 前期研究显示银翘散能够提高流感病毒感染小鼠脾脏自然杀伤细胞(NK)活性;调节NK细胞相关抗病毒细胞因子的血清含量;降低肺脏炎症因子p65mRNA的表达(p65是NF-κB信号通路活化的关键蛋白)。本课题拟进一步从流感病毒的免疫逃逸与NK细胞的免疫监视的理论视角,探讨银翘散对流感病毒感染NK细胞免疫学功能的影响。课题以银翘散作为研究对象,通过传统中医药君、臣、佐、使的拆方研究模式,结合中药化学理论研究模式和数字设计模式的研究方法,考察银翘散及其拆方对流感病毒感染小鼠肺炎模型的治疗作用及对肺脏NK细胞表面活化抗原表达的影响;并进一步建立流感病毒感染NK细胞模型,采用分子生物学方法,检测流感病毒感染过程中,NK细胞介导的抗体依赖的细胞毒作用、天然细胞毒作用以及细胞凋亡作用中的关键分子,并检测NK细胞中流感病毒载量,阐明银翘散对流感病毒感染NK细胞免疫监视作用调节的药效物质基础。

英文摘要: The previous studies have shown that Yinqiaosan recipes improve the natural killer (NK) cell function in spleen of influenza virus-infected mice, regulate the serum concentrations of antiviral cytokines related with cytotoxicity of NK cells, and reduce the p65 mRNA expression in lung. p65 is a critical inflammatory cytokine when NF-κB signaling pathway is activated. This project intends to further study the molecular mechanisms of immune surveillance role on influenza virus-infected natural killer cells medicated with Yinqiaosan recipes. Yinqiaosan recipes will be used in the study and will be decomposed according to the principles of formulating prescription in traditional Chinese medicine combined with natural pharmaceutical chemistry theory and factorial analysis method. Treatment of Yinqiaosan recipes and its decomposed recipes on influenza virus-infected mice will be explored; respectively. Simultaneously, the activated receptors on pulmonary NK cells will be detected in pneumonia mice. Influenza virus-infected NK cells will be used as a model in the study. Antibody-dependent NK cell-mediated cytotoxicity, innate cytolytic effects, apoptosis effects induced by TNF-α and effect on viral replication will be examined in the treatment of Yinqiaosan recipes and its decomposed recipes; respectively. Comparison will be conducted in the medication between Yinqiaosan recipe and its decomposed recipes, and the pharmacodynamic material will be elucidated in this study.
