中文摘要: 本课题拟利用金属卟啉配合物具有很好的生物相容性以及易于和氨基酸、蛋白质形成超分子的性质,将该配合物通过化学方法接枝到自制的多孔聚合物材料上,制备对组氨酸蛋白质具有分子识别能力的聚合物整体柱,作为高效液相色谱的固定相,与Agilent MARS Human-6 免疫亲和色谱系统组成二维液相色谱系统,去除人血浆中的高、中丰度蛋白,并通过分子识别机制富集组氨酸蛋白质,然后通过质谱对富集得到的组氨酸蛋白质进行分析、鉴定,从而建立一种基于分子识别机制的人血浆低丰度蛋白质的多维液相色谱-质谱分离、分析方法。

英文摘要: In this topic, metalloporphyrin coordination compound is used as the molecular recognition subject compound for its good biocompatibility and it is easy to form supramolecule with proteins. The metalloporphyrin coordination compound is grafted to the prepared porous materials by chemical methods to obtain a polymeric monolith with molecular recognition for histidine protein. The post modified polymeric materials are used as the stationary phases of high performance liquid chromatography in conjunction with Agilent MARS Human-6 system to remove high and middle-abundance proteins from the human plasma and low-abundance proteins are enriched via molecular recognition mechanism by the two-dimensional liquid chromatography. Then the low- abundance histidine proteins are analyzed with MS. Herein, a method for the separation and analysis of low-abundance proteins in human plasma is established based on the mechanism of molecular recognition.
