中文摘要: 利用分子生物学技术检测食品中的有害污染物是食品安全检测领域的新趋势,核酸适体检测技术是其中的热点。本项目针对农产品中重点监控的有机磷农药,利用非固相化的适体筛选方法同时针对多个有机磷农药靶分子进行核酸适体筛选。根据筛选到适体的二级结构及相应的结合位点,设计、合成分子信标;利用分子信标和有机磷农药竞争结合适体的原理建立竞争抑制法,快速筛查有机磷农药。该方法将核酸适体和分子信标技术有机结合,具有灵敏度高、识别性强、操作简单,不必与未反应的探针分离即可实时检测的优点,检测更方便、更快捷,将为有机磷农药的快速筛查开辟新的途径。

英文摘要: The molecular biology technique is the new trend on the deleterious substance detection in food safty testing field,and aptamer technology is one of the focuses. A new method of screening aptamer without target molecules immobilized will be established, which can select aptamers against several targets molecule simultaneously. This method will be applied to the selection of aptamers binding to organophosphate pesticides in primary products. Molecular beacons will be designed based on the second structures and binding sites of the selected aptamers, which are partly complementary with the middle constant nucleotide sequence of the aptamers. A competitive inhibition method was developed and applied to screening of organophosphate based on aptsmers and molecular beacons. The new method will be potential in detection of organophpsphate pesticides owing to its excellent characteristics such as high sensitivity, strong identification, and simple operation.
