中文摘要: 蛋白质差异表达与序列变异已被证实与复杂疾病发生过程等存在显著关联。目前亟待解决的问题之一就是如何将遗传变异等信息与蛋白质表达谱整合,为复杂疾病研究提出新理论、探索新方法。基于我们关于蛋白质串联质谱鉴定、差异表达、突变识别等多方面的生物信息学研究积累,本课题拟通过整合蛋白质表达谱与序列变异信息,对影响蛋白质表达的数量性状位点pQTL提出系统的蛋白质组学鉴定算法,并在方法建立、评估与应用方面展开研究:1)改善和标准化氨基酸突变的串联质谱鉴定体系;2)基于pQTL的概念,通过整合蛋白质表达谱与序列突变信息,提出基于高通量蛋白质组学的pQTL鉴定新方法,并通过理论模拟进行方法准确性、稳定性的评估。3)在扩展性研究中,利用生物网络与pQTLs鉴定进行相互补充与验证,探究其作用机理。4)将新方法应用到癌症样本中,实现对癌症pQTLs的全蛋白质组扫描鉴定,以期找到新的关键致病位点。5)开发相关软件工具。

英文摘要: It has been found that both protein differential expression and genomic variations are significantly associated with complex diseases. Therefore by integrating proteomics data and other genetic information, developing novel theroy and approach for exploring the mechanism underneath complex diseases is emerging as an important and challenging issue now. In the preliminary studies, we have made achievements in protein identification, differential expression analysis, variation detection by shotgun proteomics. In this proposal, we seek to create new method to identify the loci (variations) that disturb the protein expression quantitative traits (pQTL) during the genesis of complex disease by shotgun proteomics. We will: (1) improve and standardize the workflow for identifying the amino acid mutations from shotgun proteomics data. (2) create a novel system for identifying pQTLs by integrating protein expression profiles and amino acid variations. And then evaluate the accuracy and robustness of the proposed method by simulation. (3) in further study, prioritize pQTLs identification and functional annotation with the assistance of biological network. (4) apply the new approach into the real proteomics data from cancer samples, and scan the proteome-wide the amino acid variation so as to reveal possible risk factor
