中文摘要: 我们前期研究证实少突胶质前体细胞(OPCs)与雪旺细胞(SCs)共移植可以更好地促进大鼠脊髓损伤(SCI)的修复,并首次发现SCs及其条件培养液(SCM)可在体内、外显著促进OPCs的存活、增殖和迁移,但其机制尚不清楚。因此,本研究拟通过蛋白芯片、双向电泳和质谱分析等技术筛选、鉴定介导SCs促进OPCs存活、增殖和迁移的关键因子,并进一步研究SCs通过这些因子促进OPCs存活、增殖和迁移的机制。在阐明上述机制的基础上,以鉴定出的关键因子或下游信号分子为靶点,直接动员内源性OPCs或联合OPCs移植治疗SCI。本研究将有助于理解SCs对OPCs发挥作用的分子机制,并有望为SCI及其它脱髓鞘疾病提供新的治疗策略。

英文摘要: Our previous study has shown that co-transplantaton of oligodendrocyte progenitor cells (OPCs) and Schwann cells (SCs) promotes rat spinal cord injury (SCI) repair, and has found for the first time that SCs and its conditioned medium (SCM) strongly promote survival, proliferation and migration of OPC in vivo and in vitro. However, the mechanism which SCs affect OPC growth remains unknown. Thus, in the present study, SC-secreted key factors which promote survival, proliferation and migration of OPCs will be identified by protein chip, two-dimensional electrophoresis and mass-spectrometric assay. Moreover, the molecular events that occur during SCs-induced survival, proliferation and migration of OPCs will be explored. On the basis of the above-mentioned study,the key factors or downsream signal molecules will be applied to mobilize endogenous OPCs directly or to combine OPCs transplantation for SCI treatment. This study contributes to understanding the mechanism of SCs-induced survival, proliferation and migration of OPCs and will provide new strategy for treatment of SCI and other demyelinated-associated diseases.
