中文摘要: 生物标记在复杂疾病的临床诊断治疗和基础科学研究上都有着非常重要的地位和意义。本项目计划从系统生物学角度出发,研究针对异质性复杂疾病的动态网络生物标记识别的可计算建模与高性能算法,包括异质性疾病的多层次动态网络模型与构建算法、整合多层次生物数据的动态网络生物标记模型、以及高效的动态网络生物标记识别算法等。同时选择一些重要的复杂疾病进行动态网络生物标记的示范应用研究,例如复杂疾病预警、诊断和预后方法,复杂疾病发生发展机制研究,药物组合与旧药新用研究等。本项目将紧紧围绕重大计划的总体目标,通过对计算系统生物学中的几个重要的基础共性问题:生物医学数据的网络表示、网络比较、网络评价、子网络搜索等的高性能算法研究,开展动态网络生物标记识别的可计算建模和算法研究,为“人口与健康”这一重要科学前沿和重大国家需求中的复杂疾病研究提供关键的计算技术和数学方法支撑。

英文摘要: Biomarkers play an important role in the clinical diagnosis and therapy and the basic scientific research of complex diseases. From the viewpoint of systems biology, this project will study the computable modeling and high performance algorithms for the dynamic network biomarker identification in heterogeneous complex diseases, such as the modeling and algorithms of multi-level dynamic network construction for heterogeneous diseases, the modeling of dynamic network biomarker with integration of multi-sources biological data, and the efficient algorithms for identifying dynamic network biomarkers. Several demonstrative applications will be conducted on selected important complex diseases, for example, the methodologies for early-warning, diagnosis and prognosis of complex diseases, the development and progression of complex diseases, and drug combination and repositioning. This project will focus on the high performance algorithms for the common algorithmic problems of computational systems biology such as the network representation of bio-medical data, network comparison, network evaluation, and sub-network search. This project will provide the key computational and mathematical methodological support for the research on complex diseases.
