中文摘要: 氮肥的过量施用是设施生产上的重要课题,而维生素C(Vc)是黄瓜的主要营养品质。植物中存在四条Vc合成途径,这对氮调控Vc合成代谢的分子机制的研究带来了不小难度。我们前期的研究发现在黄瓜苗期和果期进行短期适度低氮处理,成熟果实Vc含量显著提高,且不影响单株产量。本课题拟通过早期低氮胁迫下转录组学及蛋白质组学研究,发现低氮胁迫下调控Vc合成和代谢的相关通路,并推测调控相关通路的上游调控因子。再通过生物化学、分子生物学和反向遗传学手段,进一步验证低氮调控的黄瓜Vc合成和代谢的相关通路,阐明其中低氮调节的关键步骤及控制此步骤的重要调控基因,初步构建低氮调控Vc合成的上游信号网络。深入揭示低氮处理调控Vc含量升高的分子机制,最终为改善环境和提高蔬菜品质提供理论依据。

英文摘要: Nitrogen fertilizer management research is of great theoretical and practical significance, and Vitamin C is the primary nutrient in cucumber fruit. Our previous studies have indicated that after a short-term low nitrogen supply during seedling and fruit stage respectively, Vitamin C content increase, while fruit yield show no significant difference. Previous research demonstrated that four Vc biosynthesis pathways existed in plants, which brings difficulties to study. In current research, with proteome analysis and transcriptome analysis, we attempt to found which Vc metabolism pathway regulated by low nitrogen, and speculated which regulatory factors participated in these process. Through reverse genetics, biochemistry and molecular biology methods, we will further uncover the molecular mechanism of short-term low nitrogen supply on cucumber Vitamin C induction, and find the critical process and important genes involved in this regulation. The elucidation of these problems will provide the theoretical basis for the improvement of environment and vegetable quality.
