中文摘要: 含三氟甲基的有机化合物通常具有显著增强的脂溶性和代谢稳定性。由于这些特性有助于药物在生物体内的吸收和转化进而增强药理活性,致使含三氟甲基有机化合物的合成方法研究日益受到重视,已成为目前研究的热点。在含三氟甲基化合物的合成研究中,关于三氟甲基化反应的报道不断增加。与此相比,由于alpha-三氟甲基碳负离子不易制备等原因,导致含三氟甲基中间体在合成中的应用研究明显滞后。因此,具有广泛用途的含三氟甲基合成子的研究具有实际意义、也更具挑战性。鉴此,本项目在已有工作基础上,设计了新型的含三氟甲基的多官能团1,3-和1,5-双亲电合成子。利用其具多反应位点、可以和众多1,2-及1,3-双亲核体反应等特点,建立含三氟甲基化合物的多样性、高效合成新方法。

英文摘要: CF3-containing compounds, owing to their fluorine moieties, have unique chemical and physical properties. For example, they can increase the metabolic stability or the lipophilicity, which can enhance the biological activity of a drug. For these reasons, increasing research focuses on the synthetic methods for CF3-containing compounds, especially on the trifluoromethylation reactions. On the other hand, however, the versatile synthons for the synthesis of CF3-containing compounds remain a challenge partially due to the difficulties to generate alpha-trifluoromethyl carboanion. For the importance of developing versatile CF3-containing synthons, this research proposal, based on our previous studies, is devoted to the construction of highly functionalized CF3-containing 1,3- and 1,5-dielectrophilic synthons with the aim of exploring the strategies to achieve CF3-containing compounds, for example by reaction with various 1,2- or 1,3-dinucleophiles, in an efficient and divergent manner.
