中文摘要: 干细胞是近年世界生物学界研究的热点,而循环间充质干细胞(PBMSCs)相 关研究尚少,在我国基本处于研究空白。我们通过前期大量基础实验证实,在人、大鼠、 兔体内存在PBMSCs,具多向分化潜能;比较研究证实PBMSCs 与骨髓来源间充质干细胞形态 功能类似,但在基因表达谱上有差异;在骨折愈合过程中,PBMSCs 通过血循环活跃富集于 损伤部位,可作为骨组织工程研究中干细胞的新来源。本项目拟通过旋转生物反应器建立 富集PBMSCs 的培养条件和方法;采用基因过表达方法,通过慢病毒载体系统稳定转染前期 已鉴定出的上调最明显的基因,研究其调节细胞增殖、分化和动员的作用;进行动物体内 植入实验检测其成骨潜能以及对骨折愈合的影响。通过本项目研究,将进一步拓宽对PBMSCs 的认识,建立富集PBMSCs 新方法,研究调控间充质干细胞在组织修复和再生中新的重要功 能基因,进而为干细胞和骨组织工程的应用引入新的细胞或基因治疗手段。

英文摘要: There are few studies on peripheral blood derived mesenchymal stem cells (PBMSCs). We and others have demonstrated that PBMSCs existed in human, rabbits and rats, and are multi-potent stem cells. Although there are similarities between PBMSCs and bone marrow derived MSCs, there are differences in their gene expression profiles. We have demonstrated that PBMSCs may be recruited into the sites of skeletal injury to participate in healing, which may be a novel alternative cell source for skeletal tissue repair and regeneration. This project will examine the methods for isolation, culture, and enrichment of these circulating stem cells and the potential key genes that govern their functions and homing capacities. This study will broaden our understanding of how stem cells are mobilized and released into the peripheral circulation and homed to sites where they are needed to heal damaged tissues.
