中文摘要: 研究表明miRNAs表达与肿瘤发生存在着密切关系,血液中miRNAs表达谱的稳定性,高度保守性、时序性和组织特异性使其成为一类理想的肿瘤标记物。最近本课题组发展了一种双链特异性核酸酶(DSN)信号放大技术,并建立了多重miRNA检测方法。在此基础上,本项目将进一步研究DSN酶催化机理及效率等基础理论,有效提高DSN酶介导的miRNAs检测的灵敏度和稳定性,并将DSN酶介导的信号放大机制拓展到电化学平台。同时还将建立一种miRNAs介导的等温扩增和以荧光纳米银簇为信号输出的无标记靶标介导的信号放大新方法,为高灵敏度miRNA检测新技术研究及临床诊断应用提供基础。

英文摘要: There is accumulating evidence that miRNAs expressions are frequently dysregulated in the development of a variety of cancers, and their expression patterns in cancers appear to be tissue-specific. miRNAs profiles are accessible from blood, in a remarkably stable form. Owing to these properties, miRNAs have become an ideal class of blood-based biomarker candidates for clinically validated cancer detection. Recently our group have developed a signal amplification strategy of duplex-specific nuclease signal amplification (DSNSA), and applied it in multiplexed miRNAs detection. In this project, we will further study the basic theory of the DSN's enzymatic mechanism and efficiency, and effectively improve the sensitivity and stability of the DSN method. Moreover, we will extend the DSN-aided signal amplification mechanism to the electrochemical platform. We will also establish a new miRNAs detection method based on miRNAs-mediated isothermal amplification and fluorescent nano-silver clusters as the signal output, which will provide a theoretical basis for the sensitive miRNAs detection in new technology development and clinical diagnostic applications.
