中文摘要: 花色苷是山葡萄所含有的一类重要多酚类物质,是次生代谢产物,可以吸收紫外线,对果实色泽、风味、口感和营养价值等都具有重要的作用。花色苷在生理环境下表现出强烈的抗氧化作用、防癌抗癌、具免疫调节活性、可保护肝脏和降胆固醇等,其药用价值极高。 山葡萄是我国野生果树的重要资源,果皮中花色苷含量高。深入研究参与花色苷生物合成的相关酶,对了解花色苷在山葡萄果实中的合成与分布,进一步对其进行调控具有重要意义。本研究针对山葡萄遗传资源的特异性,对山葡萄果皮转色机理进行深入探讨;对山葡萄果皮花色苷生物合成相关结构基因(F3H、F3′H和F3′5′H等)和转录调节因子(MYB和MYC等)进行克隆表达研究;了解花色苷生物合成与各基因表达的关系;研究相关基因的功能,各种相关酶的酶学反应特性;构建植物表达载体,进行转基因工作研究,为有效利用我国山葡萄基因资源建立基础。

英文摘要: Anthocyanins are a kind of important polyphenol secondary metabolites in Vitis amurensis, and which can absorb ultraviolet radiation, play an important role to the fruit color, flavor, taste and nutritional value. Anthocyanins have high medicinal value and show strong antioxidant, anticancer, immunomodulatory activity, protecting liver and lowering cholesterol at physiological environments. V. amurensis is an important wild fruit resource, anthocyanin content is highly in skin. Thorough research the related enzymes which involved in anthocyanin biosynthesis have great significance to understanding the synthesis and distribution of anthocyanins and its further regulation in V. amurensis fruit. Aimed at the study of genetic resources specificity, we should make a thorough discussion for color-changing mechanism in skin; cloning expression to the related structural genes (F3H, F3 ' H and F3' 5 ' H et al.) and transcription regulator (MYB, MYC et al.) of V. amurensis skin anthocyanin biosynthesis; understanding the relationship between biosynthesis and gene expression; the function of genes, enzymatic reaction characteristics of various related enzymes; construction of plant expression vector, transgenic research, providing basis on the efficient utilization of genetic resources of V. amurensis.
