中文摘要: 肥胖症是哈萨克族人群的高发病,课题组前期发现肠道主要菌群比例失衡在哈族儿童发生肥胖中发挥作用,但其通过何种途径影响肥胖发生尚不清楚,拟通过建立肠道菌群人源化模型进一步研究其作用机制。代谢组学分析方法可考察生物体系受刺激或扰动前后代谢产物的动态变化,研究生物体系的代谢网络,发现异常代谢物,有助于探索疾病的病理机制、预防、临床诊断等。为研究由哈萨克族肥胖儿童粪便建立的肠道菌群人源化模型鼠体内代谢特点及可能的生物标记物,本研究选用无菌鼠建立肠道菌群人源化模型,利用GC-MS技术的代谢组学分析方法,对肠道菌群人源化模型鼠血清、尿液进行代谢指纹分析,获取有效的信息以及筛选、鉴定未知生物标记物,用于研究人源化模型鼠模型的物质能量代谢特点,寻找差异代谢物,探讨哈萨克族肥胖儿童肠道菌群改变对人源化动物体内代谢的影响,以对儿童肥胖的发病机理有更深入的了解。

英文摘要: The prevalence of obesity in Kazakh is high than others. An association between reduced gut Bacteroidetes and Bact/Firm ratio with obesity in Kazakh children was identified in our former study. Further studies are necessary to elucidate the mechanism behind these changes as well as the value of determining their presence for predicting obesity. Metabonomic technology could analyse metabilites and metabolic intermediates in the biofluid qualitativly and quantitatively to discover dysbolism in the disease process, which not only help us understand the metabolic pathways of the disease process and substance of the body better, but also contribute to the discovery of disease biomarkers and supporting clinical diagnostic purposes. In this study, in order to studying human flora associated animal caused obesity-resistant material and energy metabolism of rats by analyzing serum and urine metabolic fingerprint ,screening biological markers, we set up the human flora associated animal and constructed an analysis by GC-MS-based metabolomics. After the serum sample of urine by pre-treatment, in order to looking for differences in metabolites, a better understanding of the pathogenesis of obesity and other metabolic diseases, The data collected after the peak matching, normalized correction,orthogonal signal correction (OSC) was used to multivariate analysis.
