中文摘要: DNA以及与DNA相互作用的化学和生物功能性分子在生命活动中起到了非常重要的作用,也衍生出多种结合和反应事件。申请人实验室发展了一种DNA序列信息转换成蛋白阅读框内的序列信息、并且通过生物体内的精确翻译表达成可观察的表型信号的方法,并成功的将这种方法用于DNA检测领域,初步证明了诊断与DNA有关的事件的可行性。申请人希望利用这种方法进一步实现与DNA相互作用蛋白的结合性监测,与DNA相互作用的酶类的反应性监测,与DNA相关的小分子抗癌药物的筛选以及蛋白与蛋白相互作用的研究,从而为深入研究DNA相关事件及蛋白功能提供可靠的依据。总之,申请人希望利用这种DNA的信号转换模式研究DNA与其他生物大分子或者化学小分子的相互作用体系,并最终将其发展成为一种对各种生物大分子通用的结合性、反应性检测方法。为功能基因组学、蛋白质组学的深入研究提供更多有效的信息,为阐明人类相关疾病的分子机制服务。

英文摘要: DNA and DNA-interacting, chemically and biologically functional molecules play an important role in all activities of living organisms. Many binding and reaction events are derived from these structures. An in-frame protein expression method for DNA diagnostics has been developed in my laboratory. The method takes advantage of the precise transcription of target DNA sequence information into in-frame plasmid sequence information and subsequent translation of plasmid information into observable, protein-based phenotypic signal. In addition, other DNA-related events can be monitored with this method. I propose the extension of the method to the monitoring of DNA-protein binding, DNA enzyme reactivities, screening of DNA-related anti-cancer small molecules, and investigations of protein-protein interactions. These studies will pave the way for a further understanding of protein functions and DNA-related events. Taken together, I hope that the signal transduction method proposed herein can be generally used in the examination of interaction patterns between DNA and other biomacromolecules and small molecules. The eventual goal is to develop a generic method for the diagnostics of binding and reactivity events from varied biomacromolecules.
