中文摘要: "化皮病"是仿刺参(Apostichopus japonicus)在室内及池塘养殖期间的主要流行病。研究表明,灿烂弧菌(Vibrio splendidus)等多种病原菌都能引起仿刺参的"化皮病"。仿刺参"化皮"过程的免疫应答是一个多基因调控的、多因素参与的、动态的复杂过程。深入研究"化皮"过程中差异表达基因以及这些基因之间的相互作用,是仿刺参免疫防御机制研究的基础。本研究拟通过RNA-Seq对仿刺参"化皮"不同阶段进行转录组测序和动态表达谱分析,筛选仿刺参"化皮"过程中的差异表达基因(mRNA)和差异miRNA,研究这些差异表达基因作用的信号通路;分析差异基因和miRNA动态表达模式与"化皮"程度的关系;通过预测差异miRNA的靶基因,以及对仿刺参"化皮"各阶段的转录组差异mRNA和miRNA 进行对接分析,构建miRNA-靶基因调控网络,为病原菌引起的仿刺参"化皮病"的防控提供科学依据。

英文摘要: "Skin ulceration" is a main epidemic disease in the indoor and outdoor culture of sea cucumber Apostichopus japonicus. Some studies indicated that many bacteria pathogen, such as Vibrio splendidus, can induce the "skin ulceration". The immune response during "skin ulceration" is a complicated and dynamic procedure affected by many factors and is regulated by multiple genes. To study these differentially expressed genes and their relationship in "skin ulceration" is necessary to learn the defense mechanism of A. japonicus. In this study, we plan to sequence the transcriptome of A. japonicas by RNA-Seq, and detect the differentially expressed mRNA and miRNA between the healthy individuals and "skin ulceration" individuals at different infected stages. In addition,we will study the signal pathway of these differentially expressed genes, and analyze the correlation between the dynamic expression patterns of these genes and "skin ulceration". Furthermore, we will construct the miRNA-target genes regulatory network by docking analysis of the differentially expressed mRNA and miRNA. Our study will be useful to the disease prevention of sea cucumber "skin ulceration"caused by bacteria pathogen.
