中文摘要: 二氧化碳伤害是果蔬贮藏过程中常见的生理病害,已成为引起果蔬采后损失的重要原因之一。本项目拟采用鸭梨果实为试材,围绕呼吸代谢这一关键环节,利用代谢组学的手段,研究呼吸代谢途径中重要中间产物组成和含量的动态变化规律,分析二氧化碳伤害果实具有的代谢指纹图谱,研究二氧化碳对鸭梨果实呼吸代谢网络的调控机制;通过观察呼吸代谢关键酶的变化规律,阐述二氧化碳对果实呼吸代谢途径的影响;通过观察细胞产生能荷的变化,探讨能量代谢在二氧化碳伤害发生中的作用;通过研究二氧化碳对呼吸链电子转移关键酶的影响,揭示活性氧的产生机制;利用生物显微技术,对活性氧进行亚细胞定位,阐明高浓度二氧化碳下果实主要活性氧物质的组织分布规律。研究结果将为揭示鸭梨果实二氧化碳伤害的发生机制提供重要的理论依据,为二氧化碳伤害的预测和进一步寻找二氧化碳伤害的调控手段奠定基础,为发展鸭梨的精准贮藏技术提供理论支持。

英文摘要: Carbon dioxide (CO2) injury is a common physiological disorder during the storage of fruits and vegetables and becoming one of the major limiting factors of fruit quality. The main goal of this project is to reveal the mechanism of CO2 injury. In this project, metabolomic technology will be used to investigate the effect of CO2 on respiratory metabolic intermediate products and on respiratory metabolism network of Yali pears. The metabolic fingerprint will be also analyzed. And, all of these will surround the respiratory metabolism as a key point. The influence of CO2 on respiratory metabolism pathway of fruit will be demonstrated by observing and studying the changes of key enzymes of respiratory metabolism. Meanwhile, the effect of CO2 on energy metabolism will be discussed by observing the changes of energy charge. Finally, the production mechanism of active oxygen will be revealed by studying the influence of CO2 on electron transport chain, and the distribution of major active oxygen species will be clarified. The research result will present an important theoretical basis for revealing the mechanism of CO2 injury, and it will lay a foundation for predicting the occurrence of CO2 injury and searching an effective approach to prevent CO2 injury.
