中文摘要: 本项目利用现有的广州市职业性噪声接触人群的健康监护平台,拟通过对职业噪声性听力损失相关TRIOBP基因SNP的筛查,了解TRIOBP基因miRNA结合位点SNP的分布情况;再建立TRIOBP基因多态性耳蜗毛细胞模型,通过研究毛细胞氧化损伤时TRIOBP基因多态性与miRNA(如miRNA-18a、miRNA-18b、miRNA-4735-3p等)结合的调控情况,探讨其调控机制。 本项目完成后可系统了解我国汉族职业噪声暴露人群中TRIOBP基因SNP的分布,从分子水平了解TRIOBP基因通过miRNA对毛细胞功能的调控机制,进一步阐明噪声性听力损失的遗传分子机制,筛选噪声性听力损失易感标志物,并推荐可应用的分子标记物2~3个。

英文摘要: Our planned study is designed to characterize single nucleotide polymorphism (SNP) in miRNA binding sites of TRIOBP gene among population exposed to occupational noise in Guangzhou. Moreover, we will employ a hair cell model of TRIOBP gene polymorphism to explore the molecular mechanism of TRIOBP gene was regulated by miRNA(miRNA-18a, miRNA-18b, miRNA-4735-3p, etc)under oxidative stress. Our planned studies will provide the first identification of association between SNP in TRIOBP gene and population exposed to occupational noise in Chinese Han. These studies will help us to understand the important role of TRIOBP gene in NIHL and may thereby lead to a better understanding of the molecular mechanism of NIHL. Hence, our findings may provide new insights into understanding the regulation of miRNAs on TRIOBP gene in NIHL.
