中文摘要: 体细胞重编程过程是细胞表观遗传状态剧烈转变的过程,理解重编程过程的表观遗传调控机制有重要的科学意义。我们前期工作发现H3K9甲基化是体细胞重编程过程中重要的表观遗传障碍,揭示维生素C参与了体细胞重编程的表观遗传调控机制,在本重大研究计划的集成方向上有较好的工作基础并做出了良好的成果。本项目拟延续我们对体细胞重编程表观遗传调控的研究,解析体细胞重编程驱动异染色质松散解离的调控机制,阐明异染色质松散解离过程在体细胞重编程中的作用,以此为基础筛选一系列通过松散解离异染色质提高体细胞重编程效率和质量的新因子,并进一步研究异染色质松散解离调控和重编程过程的多能性建立联系的精细分子机制。该项目直接面向本重大研究计划的总体计划,将帮助我们深入理解体细胞重编程分子机理和表观遗传调控机制。

英文摘要: Somatic reprogramming is a significant switch of cellular epigenetic state. It is important to understand the epigenetic regulation mechanism during somatic reprogramming. We previously found that histone H3K9 methylation is an important epigenetic barrier of somatic reprogramming, and studied the epigenetic regulation function of vitamin C during reprogramming. Therefore, we have proper work foundation and finished some well-known studies on the integrated direction of this major research plan. In this proposal we plan to continue researching epigenetic regulation in somatic reprogramming, including to investigate the function of heterochromatin remodeling process on somatic reprogramming and to screen a series of new factors which could improve reprogramming efficiency and quality through regulating heterochromatin dynamics. Our final aim is to understand the insight mechanism of how heterochromatin remodeling helps pluripotency reestablishment during somatic reprogramming. This project has consistent goal with this major research plan and it will benefit our understanding towards mechanism of somatic reprogramming and epigenetic regulation.
