中文摘要: 糖尿病患者作为干眼病的易患人群,仅停留在对症治疗水平上,其确切机制尚未阐明。糖尿病患者的结膜上皮细胞TGF-β和NF-kB在炎症及凋亡过程中起重要作用,我们前期研究发现糖尿病患者的泪液中CCR4+CD4+, CCR5+CD4+,CCR6+CD4的表达较正常对照组明显增多。本课题拟建立NOD.B10.H2b小鼠的蒸发过强型干眼模型,采用多层次(整体、动物模型、细胞、分子)和多方法(分子生物学、流式细胞学、激光共聚焦显微镜等)相结合的方法,检测NOD小鼠的结膜上皮细胞中相关炎症因子和凋亡因子的mRNA及蛋白表达,探讨糖尿病诱发干眼病的发病机理及其信号传导途径,为有效防治糖尿病干眼病提供新思路和新靶点。

英文摘要: Diabetic patients are susceptible persons of the dry eye and only stay in symptomatic treatment, but the exact pathogenesis is not yet clear。Our previous research have found that TGF-β and NF-kB play an important part in the inflammation and apoptosis of conjunctival epithelium of diabetes patients and observed expression of CCR4+CD4+,CCR5+CD4+, and CCR6+CD4+ cells increasing in the tear film and ocular surface in diabetic patients。This subject intends to establish the NOD.B10.H2b mice model of evaporative dry eye, which are evaluated for inflammation- and apoptosis-related cytokines mRNA and protein expression by molecular biology, flow cytometry and the laser scanning confocal microscope。Correlation and causality between apoptosis and immune-based ocular surface inflammation by NOD mice will be studied to clarify mechanisms and signaling pathways of diabetes induced dry eye, subjecting to provide new drug targets and strategies for the treatment。
