中文摘要: 单纯性细支气管肺泡癌细胞沿着肺泡壁生长,没有间质、血管、淋巴管以及胸膜侵犯,混合性细支气管肺泡癌可见肿瘤细胞有局部外侵或转移。肺泡癌是腺癌的特殊类型,有学者认为其不应归类为腺癌。目前认为混合性肺泡癌是一种含有肺泡癌成分的腺癌。单纯性肺泡癌的预后和对分子靶向药物的反应明显优于混合性肺泡癌。混合性肺泡癌是由单纯性肺泡癌进展而来,但并不是所有的单纯性肺泡癌都进展为混合性肺泡癌,目前不清楚是那种机制导致一些单纯性肺泡癌向混合性肺泡癌进展。我们假定单纯性肺泡癌向混合性肺泡癌进展时细胞内存在着某些信号通路的异常,采用基因组学的方法,检测两种肿瘤差异的信号蛋白表达,研究单纯性肺泡癌进展的信号传导通路异常;并在体外进一步验证该信号通路对肿瘤细胞活性的影响;力争在肺泡癌以及腺癌的治疗中,找到新的靶点。我们的研究发现在肺泡癌的进展中存在骨桥蛋白-整合素信号通路、CLDN1/18信号通路、ATF2信号通路、胰岛素样生长因子2信号通路的异常,这些异常的信号蛋白可能作为肺腺癌治疗的新的靶点。

英文摘要: BAC is lung carcinoma in situ. cancer cells of BAC show lepidic growth without evidence of stromal, vascular and pleural invasion.Tumors with BAC featurs and areas of invasion are classified as adenocarcinomas of mixed subtype(mixed BAC ). The prognosis and reaction to targeted drugs are better for BAC compard to mixed type lung adenocarcinoma. The mixed BAC develops from pure BAC, but not all pure BAC progresses to mixed BAC. we didn't know the mechanism of some BAC developing to mixed BAC. we postulated that there are some signal transduction pathways which are associated with the developing progress.we tested these signal transduction pathways using genome method.We found abnormal expression of the osteopontin-integrin, CLDN1/18,ATF2, Insurlin-growth factor 2 signal pathway.These moleculars may become the therapy target for the treatment of lung adenocarcinoma in the future.
