中文摘要: 苹果果实的色泽表型是由品种的基因型决定的,是品种固有的特性。然而,前期研究发现在渭北黄土高原地区的非红色苹果品种在套袋处理后呈红色。本项目在前期研究的基础上,以金冠、澳洲青苹和浓冠为材料,首先分析MYB1启动子甲基化修饰区域,通过构建启动子缺失载体并转化番茄分析甲基化区域的生物学功能;其次分析光胁迫程度、MYB1基因启动子甲基化、MYB1基因表达和花青苷合成相互之间的调控关系,鉴定MYB1启动子甲基化在调控光胁迫诱导的果实着色中的关键作用;并利用甲基转移酶抑制剂5-杂氮-脱氧胞苷处理果实,进一步验证去甲基化作用对花青苷合成的调控作用;通过分析不同色泽类型苹果品种和杂交群体中MYB1启动子的甲基化水平,鉴定MYB1启动子甲基化与苹果果实色泽遗传变异的关系。以期从表观遗传学水平解析光胁迫如何调控苹果果实中花青苷合成基因的表达,为通过育种或栽培的手段提高苹果果实的色泽品质提供指导。

英文摘要: Apple skin color is determined by the genotype of the cultivar, which are the inherent characteristics of species. However, the peel turns red due to anthocyanin accumulation after the removal of a bagging treatment in the Loess Plateau region. In this proposal, Golden Delicious , Granny Smith and Nongguan will be used as materials to carry out the projects as follows. The methylated region in the MYB1 promoter will be determined firstly. The biological function of this region will be surveyed through transformation of the constructs with the fragment-deleted promoter. The relationship among the photo-stress,MYB1 promoter methylation,the expression of MYB1 gene and the synthesis of anthocyanins is investigated. The regulatory function of the MYB1 promoter methylation is analyzed. The effect of the de-methylation on the synthesis of anthocyanins is ivestigated by application of 5-AZA-dC, a kind of inhibitor of methyl transferase. The correlation between the DNA methylation of MYB1 promoter and the variation of apple skin color will test. The mechanism of the anthocyanin synthesis in the peel of non-red apple cultivar regulated by photo-stress will be investigated from DNA methylation, providing a base for improving the behavior of coloration by means of breeding and cultivation.
