中文摘要: 炎症性肠病是一组原因不明的慢性肠道炎症性疾病,其确切病因及病机尚未完全明确,为临床有效治疗带来一定困难。本课题组经近十年的临床经验总结,结合炎症性肠病的湿毒内蕴、气血壅滞、脾肾亏虚病机,创立红萸饮,以清热解毒、凉血活血,兼之温补脾肾,在临床上取得良好疗效。本研究在前期研究基础上,拟从三方面证实IL-6/STAT3信号通路与IBD发病的关系。采用体外细胞培养技术,研究红萸饮拆方对JAK2/STAT3信号通路的影响作用;采用组织病理学技术与方法,研究炎症性肠病(IBD)造模小鼠及IBD患者结肠黏膜观察肠黏膜损伤情况;采用分子生物学、免疫组化等技术与方法,检测血清及肠粘膜IL-6和STAT3、磷酸化STAT3蛋白及其下游基因c-myc、surviv等的表达。证明IL-6/STAT3信号通路与IBD的关系,并且中药红萸饮通过抑制该信号通路而起到治疗作用。

英文摘要: Inflammatory bowel disease is a group of unknown cause chronic inflammatory bowel disease, its exact etiology and pathogenesis has not yet fully understood, and it is difficulty to cure. In the course of the nearly 10 years of clinical experience, we created Chinese decoction Hongyuyin under the theory of chinese traditional medicine to heat-clearing and detoxifying, cool blood ,promoting blood circulation, together with warm reinforcing spleen and kidney,and have good affect in clinical. This study based on the previous studies, from three aspects to study the relationship between IL-6 / STAT3 signal pathway and IBD. We plan to research Hongyuyin recipe and its disassembled prescriptions and its function in IL-6 / STAT3 signaling pathways with cell culture.We observe the colonic mucosa injury in mice and IBD patients with the histopathological technology and method.Using molecular biology, immunohistochemical techniques and methods, we study the amout of IL-6 and STAT3, pSTAT3 and downstream gene protein c-myc, surviv and expression. We want to prove the ralationship between IL-6 / STAT3 signaling pathways and IBD, and Hongyuyin can inhibiting this signaling pathways and have therapeutic effect.
