中文摘要: MicroRNAs(miRNAs)是一类广泛存在于真核生物体内、长约20-25nt的非编码小分子RNA,以序列特异性方式调节基因表达。研究显示,miRNA与原发性高血压发生有一定相关性,其分子机制仍有待进一步研究。非洲绿猴是唯一具有原发性高血压特征的非人灵长类动物,其血管内皮细胞对高血压有一定适应性和保护作用,但相应高血压形成及血管保护的分子机制尚不明确。本研究拟对非洲绿猴血管内皮细胞miRNA表达谱进行分析,筛选出非洲绿猴差异表达miRNA分子以及其新靶基因,并且从细胞模型上进行miRNA功能研究。同时,本研究拟对非洲绿猴miRNA单核苷酸多态性(SNP)特征进行分析,以期阐明非洲绿猴自发性高血压形成及血管保护的分子机制,为高血压发病机制研究及其防控提供新策略。

英文摘要: MicroRNAs (miRNAs) represent a highly-conserved novel class of small endogenous non-coding RNAs with 20-25nt which can negatively regulate gene expression. Recent studies demonstrated that miRNAs are involved in cardiac hypertrophy and heart failure. However, little is known about the roles of miRNAs in essential hypertension. Single nucleotide polymorphisms (SNP) in miRNA are recently proved to be related to several diseases. African green monkeys (AGM) are the only non-human primates with essential hypertension, of which the molecular mechanism is unclear. This study aims to investigate both the plasma and endothelial cell miRNA expression profiles of AGM, as well as the miRNA SNP location and its functions.
