中文摘要: 细胞内质网应激-自噬途径在宿主的感染、免疫及各种疾病中都发挥着十分重要的作用。近年来,内质网应激-自噬与疾病的关系是生命科学、医学研究的热点之一,而在畜牧兽医方面的研究不多,对细胞内质网应激-自噬与病毒感染、动物疫病的关系及作用所知甚少。鉴于此,项目根据内质网应激-自噬的最新研究成果,针对猪瘟发病机制不清楚、病毒免疫逃逸等这些关键科学问题,结合猪瘟发病特点,在我们前期工作基础上,以细胞内质网应激-自噬途径在猪瘟病毒感染与免疫调控中的机制为研究主线,从猪瘟病毒体内/外感染入手,选择相关细胞、内质网应激分子、自噬分子、凋亡因子及细胞因子等为研究对象,结合内质网应激/自噬调节剂、基因干预等手段,从不同层次和角度进行研究,以揭示细胞内质网应激-自噬对猪瘟病毒感染、免疫调控的分子机制,使我们更加明确猪瘟病毒感染对机体的损伤新机制, 进而从新的角度了解猪瘟的发病机制,为猪瘟的防控寻找新途径、提供新策略

英文摘要: Cell endoplasmic reticulum (ER) stress-mediated autophagy, as a defense mechanism of the body, plays a very important role in host infection, inflammation, occurrence and development of various kinds of diseases, as well as the immunological process. In recent years, the relationship between cell ER stress-mediated autophagy and the diseases has become a hot spot in life science and medical research. However, few studies related to cell ER stress-mediated autophagy have been done in animal husbandry and veterinary. Furthermore, correlation and roles of cell ER stress-mediated autophagy in animal diseases, immunological regulation and viral infection are poorly understood. So, based on our previous studies and according to the theory of ER stress-mediated autophagy, the latest findings and the features of CSF development, this project aims to reveal molecular mechanisms of pathogenicity and immune regulation of cell ER stress-mediated autophagy in CSFV infection. After virus infection in vivo and in vitro, the relevant cells, molecular marker of ER stress and autophagy, apoptotic factors and cytokines are investigated using ER stress and autophagy agonists/inhibitors or gene interference. In this study, the molecular mechanisms of cell ER stress-mediated autophagy in immune regulation after CSFV infection are fully investigated from different levels and angles. It clearly indicates a new mechanism of the damage of CSFV infection to the host body, making us to understand the pathogenesis of CSF in a new perspective, bringing a new way to enhance the immune effects of CSF, and providing new strategies and new ideas for prevention and control of CSF.
