中文摘要: 肿瘤是危害人类健康的重大疾病,发展新的诊疗策略以改善肿瘤的治疗现状已经成为具有实际意义的重要需求。光热和光动力疗法是一类以光作为驱动力的治疗肿瘤的方法,由于其具有良好的治疗效果和较低的毒副作用,因而在癌症治疗方面展现出很好的应用前景。然而,如何将光热和光动力治疗进行有效结合并实现高效的靶向治疗仍然是目前面临的挑战。本项目即利用核酸适配体的优异识别性能和纳米材料在肿瘤治疗中的优势,以DNA作为分子工具,以金纳米棒和光敏二氧化硅纳米颗粒作为结构单元构建多功能纳米组装体,通过DNA连接并调控组装体的结构,实现多功能诊疗一体化,达到光热/光动力联合高效治疗恶性肿瘤的目的。通过对多功能纳米组装体在细胞以及活体小动物水平上治疗效果的研究以期为临床研究提供基础实验数据,为有效治疗恶性肿瘤提供新的策略。

英文摘要: The development of new theranostic strategies for cancer therapy is of great importance in cancer treatment. The combination of diagnosis and therapy into one single system would significantly improve the treatment efficiency. Photothermal therapy and photodynamic therapy are the light-driven therapy for disease treatment. Due to their high efficiency and low toxicity, they show great potential in cancer treatment. However, the efficient combination of targeted phototheramal and photodynamic therapy with the diagnosis capability is still a challenge. In this project, we utilize DNA as the linker to construct the assembly of silica nanoparticle and gold nanorod modified with aptamer for targeted photothermal and photodynamic therapy. The combinational treatment and the simultaneous monitoring of therapeutic effects could significantly improve the treatment efficiency. The investigation of treatment effect on tumor cells and living animals could provide the basic foundation for further clinic research and provide new insights for efficient cancer therapy.
