中文摘要: 了解受体分子的在体表达量的变化、慢性肝损伤导致肝纤维化的程度变化及肝脏功能的变化情况是分子成像领域面临的挑战和亟待解决的科学问题。随着影像医学的飞速发展,传统的影像医学与分子生物学等多学科交叉融合,影像分子成像技术不断进步,超声分子影像学作为分子影像学的重要分支,积极研究超声分子显像将会对临床医学提供重要的检测手段及评价手段。本课题以大鼠的肝纤维化为研究模型,针对肝纤维化后肝细胞表达的ASGPR的受体变化情况,体外构建其靶向配体半乳糖化多聚赖氨酸,利用超声分子显像,实时、动态、在体评估肝脏功能的变化及肝脏纤维化程度。并与病理标本检测、磁共振弹性成像技术等检测手段进行对比研究和相关性分析,获得一种对肝纤维化的发生、发展及受体的在体表达量变化的规律的实时监测的新思路与手段。

With the rapid development of Medical Imaging, image of traditional medicine and molecular biology multidisciplinary fusion, The Imaging molecular imaging technology had advanced, the supersonic molecular imaging as an important branch of molecular imaging, The actively studying in ultrasound molecular imaging will be provide clinical medicine to an important means of detection and evaluation tools. The liver fibrosis in rats is research model in this topic, Against The changes of the ASGPR of expression in liver cells, Building the targeting ligand in vitro that is the Galactosylated-Polylysine, Assessed the changes of liver function and the degree of liver fibrosis use of real-time, dynamic, in vivoment of ultrasound molecular imaging and comparative studies and analysis with the pathological specimen testing, MRI elastography technology and so on. Obtained an new ideas and means of real-time monitoring in occurrence, development, and receptors expression on liver fibrosis.
