中文摘要: 对核爆炸的幸存者、放疗病人、辐射工作者和辐射事故的研究发现重离子辐射会引起心包炎、心肌纤维化衰弱、冠状动脉疾病、心律失常、动脉硬化等心血管并发症。在空间环境中,重离子辐照是造成宇航员健康危害的主要因素,为了降低宇航员在太空飞行中辐射造成的危害,开展对重离子的研究非常的必要。斑马鱼因其独特的优势,作为模式生物已广泛应用于心血管发育和疾病相关领域的研究。前期实验已经证实重离子辐射能够引起斑马鱼心血管发育畸形,血管发育相关基因Ephrinb2、Flk-1表达明显下调,本项目将首先深入分析Ephrinb2、Flk-1基因在辐射损伤中的作用;再以斑马鱼为材料,采用基因芯片技术筛选出辐射处理组和正常组间差异表达的心血管发育相关基因,深入研究辐射引起的心血管系统损伤的分子机制。

英文摘要: Several groups of radiation-exposed human populations have shown evidence of increased cardiovascular disease (CVD) after heavy ions radiation, including Atomic Bomb Survivors (ABS), radiotherapy patients and Chernobyl workers. These CVD include pericarditis, myocardial fibrosis, muscular dysfunction, coronary artery disease, autonomic dysfunction and conduction disturbances. In space, heavy-ion irradiation is a harmfully factor to astronauts health. In order to reduce the damage in space flight by radiation, it is necessary to research on heavy ions radiation. Zebrafish is taken as a mode organism for cardiovascular development and diseases research for it's special structure. Early research has approved that heavy ion could cause teratogenesis of zebrafish embryonic cardiovascular, the mRNA expression of cardiovascular development associated genes Ephrinb2 and Flk-1 were obvious reduced. This research will deeply understanding the role of genes Ephrinb2 and Flk-1 in irradiation injury. Screening the key genes correlate with cardiovascular teratogenesis by irradiation using gene chip technology. Investigation on the molecule mechanism of irradiation effects on cardiovascular teratogenesis.
