中文摘要: 前列腺癌严重危害男性生命健康, HIFU是治疗前列腺癌的重要方法,由于HIFU消融前列腺癌存在效率低、易残留的问题,寻找副作用小、靶向性强的新型载药纳米增效剂对HIFU有效治疗前列腺癌具有十分重要的意义。项目组前期成功制备出PLGA有机纳米粒,对HIFU治疗前列腺癌具有良好的增效作用。在此基础上,我们提出设计与制备一种兼具诊断和增效双重功能的智能型载药FC-HMONs纳米粒,该纳米粒子能够穿过肿瘤血管内皮间隙,靶向富集于前列腺癌组织,在聚焦超声作用下发生液气相变,最终实现对前列腺癌高灵敏度超声造影成像和HIFU增效治疗。项目拟开展造影/增效剂化学组成与纳米结构的调控,筛选最佳的氟碳分子和负载量,检测表面包被的有机/无机杂化介孔有机硅空心纳米粒子(HMONs),考察其生物相容性,探讨响应型超声造影成像和HIFU增效性能的精确调控。项目的开展将对前列腺癌的分子影像诊断和治疗提供一种新的模式。

英文摘要: Prostate cancer caused serious damage to the health of man. High intensity focused ultrasound (HIFU) is one of the important treatment for prostate cancer. However, current HIFU therapy is still not ideal for the limitation of complete erase of the tumor cells and relatively low therapeutic efficacy. Thus, it is considerably needed to design and fabricate a novel nano-synergistic agent, with minimum side-effect and optimal tumor-targeting ability together, to improve the therapeutic efficacy of HIFU treatment in prostate cancer for the recent. In previous study, we have fabricated a series of PLGA nanocapsules, which could efficiently enhance the HIFU ablation effect for the prostate tumor. Furthermore, in this project, we propose the novel design of a hybrid and intelligent nanotheranostic agent (FC-HMONs), for the integrated function of ultrasound contrast imaging, imaging-guided HIFU synergistic therapy, drug delivery and prostate tumor targeting. The inorganic/organic hybrid structure was applied for FC-HMONs to ensure the biocompatibility in the future clinical application. In addition, both the type and loading capacity of fluorocarbon guest molecule, which is encapsulated inside the particle as the cavitation nucleus, will be carefully varied for seeking out the optimal utility of FC-HMONs in ultrasound imaging and HIFU synergistic therapy. It is expected that this project could provide a novel and high-efficient model for the accurate diagnosis and therapy of prostate cancer.
