中文摘要: 自驱动微纳发动机在药物运输、生物传感、以及蛋白质和细胞分离等方面有重要的潜在应用前景。该项目针对现有的自驱动微纳发动机存在生物相容性差的难题,考虑到金属镁(Mg)及其与水反应的产物生物相容性好,设计制备可运载并光控释药的Mg/聚异丙基丙烯酰胺(PNIPAM)-金(Au)双面神智能微米发动机。发展Mg/PNIPAM-Au微米发动机的拓扑表面改性制备方法,研究其结构形貌的调控原理,以及和水燃料介质的化学组成(包括氢氧化镁惰性层去除剂的种类、浓度和pH值)、表面张力及粘度对微米发动机的自主运动速率、轨迹和血液相容性的影响规律;研究微米发动机的光致体积相变响应时间、载药和光控释药性能对其形貌结构和水燃料介质的依赖性。分析建立与血液相容的Mg/PNIPAM-Au智能微米发动机的设计原理、制备方法,自驱动的化学反应机理、药物运载和光控释放的控制策略,为其在生物医学方面的实际应用奠定基础。

英文摘要: Self-propelled micro-/nanomotors have promising potential applications in drug delivery, biosensors, as well as protein and cell separation etc. In this project, we propose a Mg/Poly(N-isopropylacrylamide)-Au (Mg/PNIPAM-Au) smart Janus micromotor which is capable of loading, transporting and photoresponsive releasing drug molecules. In consideration of the biocompatibility of magnesium, water fuel medium and the resultants of magnesium-water reaction, the proposed Mg/PNIPAM-Au may have a promising biocompatibility compared to that of the existing micro-/nanomotors. The topological surface modification method will be developed for the fabrication of Mg/PNIPAM-Au Janus micromotors. The controlling protocols of the structures and morphologies of the Mg/PNIPAM-Au Janus micromotors will be studied. The influence of the structures and morphologies of the micromotors, as well as the viscosity, surfaces tension and compositions of the water fuel medium (including various magnesium hydroxide passive layer eliminators (MHPLEs), concentration and pH value) on the autonomous motion and hemocompatibility of the micromotors will be analyzed.
