中文摘要: 噬菌体是生物圈中数量最多的生物,在控制宿主群落演替、生物进化和地球生物化学循环中起到重要的作用。目前关于噬菌体生态学的研究,多是基于解析环境中eDNA基因序列,虽然揭示出部分噬菌体家族的基因组成及分布特征,但噬菌体与宿主间的相互关系尚不明晰,特别是针对噬菌体与大豆根瘤菌间侵染关系的研究还是空白。为此,本研究采用培养和分子生物学相结合的方法,对侵染不同基因型根瘤菌获得的噬菌体,扩增T4型噬菌体的结构基因g23进行系统进化分析,并采用脉冲场凝胶电泳PFGE和限制片段长度多态性RAPD技术分析噬菌体全基因组大小和多样性,同时利用盆栽试验接种根瘤菌和噬菌体,明确噬菌体对大豆结瘤、固氮及产量的影响。该研究将明确侵染大豆根瘤菌的噬菌体类群,揭示大豆根瘤菌噬菌体的遗传基因特征,解析噬菌体与根瘤菌间遗传基因多样性的关系,为东北大豆根瘤菌遗传资源演化及资源保护提供科学依据。

英文摘要: Bacteriophages are the most abundant biological entities in the biosphere, and play important roles in regulating the succession of host communities, biological evolution and biogeochemical cycling. Recently, the researches on viral ecology going very rapidly and getting much eDNA, indicating the genetic diversity and distribution character in different environments. However, limited studies focus on the relationships between virus and bacterial host, especially between rhizobiophage and rhizobia. Therefore, in this project, the cultural methods and molecular biological technique will be used to reveal the phylogenetic diversity of rhizobiophage. In addition, the g23 which encodes the major capsid protein of T4-type bacteriophages was choosing as objective gene for their ubiquitous and more abundant in different environment, and the length and diversity characteristics of rhizobiophage will be analysed by pulsed field gel electrophoresis and random amplified polymorphic DNA methods. Meanwhile, the effect of rhizobiophage on the nodulation, nitrogen fixation and yield of soybean will be studied by pot experiment.
