中文摘要: 肝癌是癌症中常见而且死亡率很高的重大疾病, 开展肝癌的早期诊断和治疗意义重大。本项目拟发展一系列具有高选择性与高灵敏度新型功能纳米探针,用于细胞水平上肝癌的靶向诊治和荧光成像分析。基于纳米金、纳米金属氧化物两种纳米单元,制备得到集光、磁、热、催化等特性于一体的异质结构纳米复合材料;通过连接多种肝癌肿瘤标志物的靶向识别基团,构建功能纳米探针实现多组分的原位实时检测;利用纳米复合材料催化产生活性氧自由基诱导细胞凋亡的特性以及磁疗和热疗功能,将探针自身作为一种治疗癌症的纳米药物,并开展联合疗法来特异性杀死肝癌细胞;深入研究功能纳米探针的细胞穿膜、细胞内迁移和精确定位以进一步阐明细胞凋亡和活性氧信号转导的分子机制。本项目取得的成果将为肝癌的预警和及时诊治提供新策略与新途径。

英文摘要: Liver cancer is a common form of cancer with a higher mortality rate, making it significant to carry out early diagnosis and therapy. In this project, we plan to develop a series of highly selective and sensitive bifunctional nanoprobes for liver cancer cell molecular diagnosis, targeted therapy and imaging. Heterostructured nanomaterials, which exhibit optical properties, hyperthermic effects, magnetization and catalytic activity, are prepared based on Au and metal oxide nanocomponents; In conjunction with targeting ligands, the resulting bifunctional nanoprobes can be used as fluorescent probes for real-time detection and localization of tumor makers, and also as drugs to kill liver cancer cells through catalytic formation of ROS, magnetotherapy and hyperthermia; Cell penetrating, intracellular migration and localization of these bifunctional nanoprobes will also be investigated to improve the understanding of apoptosis and ROS signal transduction mechanisms. This research will provide new strategies for improving liver cancer early warning and timely treatment.
