中文摘要: 鱼类卵子发育是一个复杂的动态生理过程,受到内外源信号通路协同调控,其中卵子内细胞间的局部信号通讯对各类细胞的发育起着极为重要的协调作用。本课题拟以大菱鲆卵母细胞微环境中促性腺激素受体(gonadotropin receptors,GtHRs)介导的PKA信号通讯为核心,以卵母细胞、颗粒细胞和膜细胞之间互作为主线,获取GtHRs编码基因,查明其时空表达规律,阐明卵子发育过程中细胞外基质和间隙连接的变化及其对卵子内各级细胞发育的调控作用,认识GtHRs对PKA信号通讯媒介作用及其对卵子选择和闭锁影响。以上研究有助于揭示大菱鲆等海水硬骨鱼类卵母细胞局部微环境中GtHRs介导的细胞间信号通讯对卵子选择、闭锁、成熟和营养物质的转运等关键生理环节的调控作用,为完善海水养殖鱼类种苗生产技术提供理论依据。

英文摘要: Egg development in fish is under control of complex regulatory mechanisms involving both extrinsic and intrinsic signalling pathways. In the case of signalling pathways, local signal communication within egg cells, appears to play critical roles in controlling development of different cells. The effect of gonadotropin receptors(GtHRs)-mediated PKA signal communication on development of turbot eggs was evaluated and involved signalling pathways as well as oocyte, granulosa cells and theca cells interaction were investigated in oocyte microenvironment. In this study, the spatiotemporal experssion patterns of GtHRs were assessed after we obtain the coding region. In addition, the changes of gap junctions and extracellular matrix within the egg and its role in regulating cell development were evaluated, together with the underlining mechanisms. Furthermore, the effect of GtHRs-mediated PKA signal communication on egg selection and atresia were identified and characterized. These studies reveal the regulation of GtHRs-mediated intercellular signal communication on egg selection, atresia, maturation, nutrients transport and other key physiological aspects in oocyte microenvironment of turbot (Scophthalmus maximus), further elucidate internal physiological mechanism in teleosts, thus providing the theory basis for f
