中文摘要: 很多芳香烃降解菌中存在冗余降解基因(簇)并发挥着重要作用。冗余降解基因(簇)作为一种进化优势,在环境受到扰动时能够增强微生物群落的抵抗力和恢复力。对其协同作用方式和分子生态意义的研究有助于生物修复工作的开展。Pseudomonas putida ND6具有多个冗余降解基因簇,对其生物学功能的研究已有一定的基础,而且ND6菌株完成了全序列测定,环境安全性好,是很好的研究模型。本研究计划开展两部分工作。第一,以ND6菌株为材料,继续加深对冗余降解基因(簇)调控机制的研究。第二,采集芳香烃污染程度不同的土壤样本,全景式观察环境中冗余降解基因(簇)的类型,分布规律,对环境胁迫的响应机制并争取发现新的冗余降解基因(簇)。通过这些工作加深对冗余降解基因(簇)存在的分子生态学意义的理解。

英文摘要: The redundancy degradation genes and clusters exists widely in polycyclic aromatic hydrocarbons (PAHs) degradation strains and plays an important role. Microbial communities possess these redundancy genes and clusters as evolutionary advantages which could enhance their resistivity and stabilization in PAHs disturbance. The study on cooperation mechanisms and molecular ecological significances of redundancy degradation genes and clusters is helpful to bioremediation. Pseudomonas putida ND6 is a good model to study the cooperation mechanisms and molecular ecological significances of redundancy degradation genes and clusters. The reason is as follow. There is more than one kind of degradation genes and clusters in ND6 strain and we have studied on them for almost ten years. The physical roles of them had been demonstrated. Moreover, the genome and plasmids of ND6 strain had been previously sequenced and annotated. Two parts of work will be carried out in this study. One is demonstrating the regulation mechanisms of redundancy degradation genes and clusters in ND6 strain. Another is making an overall view insight on the types, distribution patterns and responses to the environmental disturbances, in order to understand the molecular ecological significance of redundancy degradation genes and clusters deeply.
