中文摘要: 糖尿病肾病是导致终末期肾脏病的最常见原因之一, 不仅给患者带来巨大的病痛和负担,也给社会带来巨大的经济压力。探寻更加有效的糖尿病肾病治疗靶点具有重大的科学意义和社会价值。本课题组近八年来对长寿基因Sirt1在肾脏的生物作用进行了探讨。系列研究结果提示Sirt1及其调节系统在糖尿病肾病的发病中起重要作用。在我们前期研究的基础上,结合当前人们对糖尿病肾病及Sirt1生物学认识的进展,本研究将测试研究假说:Sirt1活性下降与糖尿病肾病进展相关,其机制可能是通过Sirt1-HIF1a-VEGF途径影响肾小球足细胞的功能,而Sirt1的活性又受到肾小管髓攀升支粗段产生的NAD的调控。本课题将采用基因敲除,细胞选择性基因敲除模型以及细胞生物学等技术探讨Sirt1及其调节系统的作用,并注重转化的探讨。若该假说成立,Nampt/NAD/Sirt1可能成为糖尿病肾病潜在的治疗靶点。

英文摘要: Diabetic nephropathy (DN) is one of the leading cause of end stage renal disease requiring renal repalcement therapy. DN not only constitutes a health threaten to patients, but also becomes a huge burden to Society. During the past 8 years, our group examined the role of aging gene Sirt1 in the kidney. Our series studies high light a potential role of Sirt1 in DN. Based on our previous studies and the advances of Sirt1 biology, we proposed a working hypothesis of the present study: abnormal Sirt1 contributes to the development of diabetic nephropathy; the mechanism by which Sirt1 protect the kidney involves Sirt1-HIF-VEGF pathway; the activity of Sirt1 in the glomeruli is controled by adjacent tubules via providing Nampt/NMN/NAD that may be affect by metabolic disorder. In the presnet proposal, state-of-art technique including conditional knockouts will be used. The present proposal also pays attention to translation potential of the study. If the hypothesis is verified, Nampt/NAD/Sirt1 could be a potential therapeutic targets for diabetic nephropathy.
