中文摘要: 恶性肿瘤是严重威胁人类健康的疾病,肿瘤发病机制的阐明是有效防治肿瘤的基础。肿瘤细胞中TRAF2主要调控NF-κB、MAPK生存信号通路,与肿瘤的发生发展密切相关,但是TRAF2功能的调控机制尚不明确。本课题组前期研究发现核受体Nur77能够与TRAF2相互作用,参与TRAF2下游信号通路的调节。在前期研究的基础上,本项目将从分子水平、细胞水平和整体动物等多个层面,采用多种技术,围绕Nur77与TRAF2相互作用的功能和调控机制,深入研究Nur77与TRAF2在肿瘤发生发展过程中的关键作用。本项目的研究将为Nur77及TRAF2的研究提供新的视角,为肿瘤治疗提供新靶点和新策略。

英文摘要: As the Malignant tumor has been a serious threat to human health, clarifying the molecular mechanism of cancer development, has become the foundation of developing rational and effective treatment. TRAF2 as one of the key adaptors regulating NF-κB and MAPK survival signaling pathway, is important for cancer development, but how it regulates is still unknown. In previous research our group has found nuclear receptor Nur77 could interact with TRAF2 and involves in the regulation of downstream of TRAF2 signaling pathway. Based on the previous research,we will using molecular biological, cell biological methods and animal model, to study the function and mechanism of Nur77 and TRAF2 interaction, explain the key role of Nur77 and TRAF2 during the cancer development. This project will provide a new sight for the research of Nur77 and TRAF2, also a new target and strategy for cancer treatment.
