中文摘要: 近年来,痕量分析在环境保护、食品安全以及疾病诊断等与人类生活密切相关的领域中显得越来越重要。其中,肿瘤标志物的超灵敏准确检测对于肿瘤早期诊断、有效治疗的开展以及预后监测具有十分重要的临床意义。由于现有检测技术普遍存在灵敏度不高的缺陷,通过增强检测信号促进灵敏度提高的信号放大技术成为目前实现低含量生物分子超灵敏检测最重要最常用的方法。核酸酶辅助的信号放大策略是最近发展起来的一种新兴信号放大技术,具有极强的普适性和便捷性。通过使用灵活多样的核酸酶,核酸酶辅助的信号放大策略有效促进了生化分析灵敏度的提高,引起了研究者浓厚的兴趣。因此,本项目拟在核酸酶辅助的信号放大策略设计研究基础上,以核酸适体作为识别元件,发展高度灵敏和特异性检测肿瘤标志物的电化学生物传感器,以期为肿瘤的诊断和预后提供有力的技术支持,并对相关的科研工作提供指导借鉴。

英文摘要: In recent years, trace analysis is becoming more and more important in environmental protection, food safety and disease diagnosis. Among them, ultra-sensitive and accurate detection of tumor markers that can monitor the occurrence of tumors in real time is of great clinical significance in diagnosis, treatment and prognosis of tumors. As the existing detection techniques always have the problem of poor sensitivity, signal amplification strategy, which can greatly improve the detection sensitivity through the enhancement of the detection signals, has been the most common but most important method to realize the ultra-sensitive detection of low levels of biological molecules. The nuclease-assisted signal amplification strategy is a newly developed signal amplification technique with the well universality and convenience. By using varieties of nucleases, the nuclease-assisted signal amplification strategy has greatly promoted the sensitivity of biochemical analysis that has aroused more and more interests of researchers. Based on the research of the nuclease-assisted signal amplification strategy, this project will use aptamer molecules as the recognition elements to develop novel electrochemical biosensors for ultra-highly sensitive and selective detection of tumor makers.
