中文摘要: 泛素-蛋白酶体途径(ubiquitin-proteasome pathway,UPP)是目前已知的最重要的、有高度选择性的蛋白降解途径。以UPP为靶目标在疾病治疗中已成为一个研究的热点。如何利用及模仿细胞内UPP途径,高选择性地对靶标蛋白进行泛素化,进而调控与疾病发生相关蛋白的量,实现相关疾病的治疗,正在引起国际学术界的重视。鉴于aptamer的高选择性和高亲和性,从提高UPP途径的选择性和靶向性的角度出发,本项目将aptamer(靶向疾病发生相关的目标蛋白和E3泛素连接酶)引入UPP途径,在体外和细胞水平上研究aptamer诱导靶标蛋白的泛素化降解以及靶标蛋白降解后对细胞的影响,初步建立可人工调控的、高选择性、普适性的诱导蛋白泛素化降解体系。本项目的研究成果将为细胞生物学的研究提供工具,具备生物制药的潜能,也将为相关疾病的治疗提供新的方法。

英文摘要: Ubiquitin-proteasome pathway (UPP) is the most important and highly selective protein degradation pathway. UPP as a target for treatment of the disease has become a hot research field recently. How to utilize and imitate the UPP pathway in cells to recognize target protein selectively and artificially regulate the amount of target protein are attracting the attention of the international academic community. Based on high selectivity and affinity of aptamer, the project will introduce aptamer into UPP pathway to improve the selectivity of the UPP pathway. The aptamer is constituted by two parts:aptamer targeting to related protein and aptamer targeting to E3-ubiquitin ligase. We aim to establish the artificial degradation system for target protein with high selectivity and universality in vitro and in vivo by ubiquitin-proteasome pathway. The research project aim to provide a new tool to cell biology research and a new method for treatment of related diseases.
