中文摘要: 流行病学研究表明电磁辐射可能是阿尔茨海默病(Alzheimer's Disease, AD)的危险因素,但尚缺乏实验研究证据。我们的研究表明电磁辐射暴露能够引起脑神经元的氧化应激,并促进β淀粉样蛋白(β amyloid protein, Aβ)的表达,学术界一致认同Aβ在AD发病中起关键作用,并且Aβ为核心的老年斑是AD的主要病理表现之一。所以本课题立足于氧化应激在电磁辐射与Aβ之间的桥梁作用,开展电磁辐射与AD间关系研究。以不同辐射源为处理手段,以SD大鼠为研究对象,应用水迷宫和旷场实验等方法检测其行为学改变;以光镜、电镜和组织化学方法检测大脑皮质、海马等部位的形态学改变。应用RT-PCR等分子生物学方法检测氧化应激、Aβ表达等相关蛋白和基因水平的变化。以期阐明电磁辐射是否引起AD疾患及其分子机制,为电磁辐射所引起的AD疾患的预防和治疗提供理论依据。

英文摘要: Epidemiological studies show that electromagnetic radiation (EMR) may be a risk factor for Alzheimer's Disease (AD), but laboratory evidence is not sufficient. Our studies have found that electromagnetic radiation exposure can cause oxidative stress of neurons. Meanwhile oxidative stress of neurons in the brain can contribute to the amyloid beta protein's (Aβ) expression. The scholars unanimously agree with the critical role played by Aβ in AD pathogenesis.And Aβ is the core of senile plaques, which is one of the main pathological manifestations of AD. So this topic is based on the bridge of neuron oxidative stress between the electromagnetic radiation exposure and Aβ expression, to carry out the research on the relationship between electromagnetic radiation and AD. This project uses different radiation sources as treatment and use Sprague-Dawley(SD) rats as the research object. We use the water maze and open field test to detect behavioral change. We use light microscopy, electron microscopy and histochemical methods to detect morphological changes of cerebral cortex, hippocampus and other parts. By RT-PCR and other molecular biology methods, we detect oxidative stress of Aβ-related protein expression and gene-level changes.
