中文摘要: 玉米淀粉合成由一系列酶催化完成的,其中二磷酸腺苷葡萄糖焦磷酸化酶(AGPase)是玉米淀粉合成的关键酶限速酶。其活性高低直接决定玉米淀粉的产量。研究表明磷酸化调节可能是AGPase活性的关键调节方式,但如何影响其活性和淀粉的合成还不清楚。本申请以西南地区广泛应用的高淀粉骨干自交系08641为研究对象,综合运用生化与分子生物学、磷酸化蛋白质组学、酶动力学方法,研究淀粉合成过程中,AGPase磷酸化对其活性影响及在淀粉合成过程中的变化规律。为靶向改造AGPase酶提供理论依据及骨干自交系08-641进一步分子改进奠定基础。

英文摘要: Maize starch synthesis is completed by a series of enzymatic catalysis, of which ADP-glucose pyrophosphorylase (AGPase) is a key rate-limiting enzyme in starch synthesis in maize. AGPase enzyme activity directly determines the yield of maize starch. Studies show that phosphorylation of AGPase may be a key regulation to its activity, however, how to effect its activity and synthesis of starch is still unknown. Our proposal is aimed to invistigate the effect of AGPase phosphorylation on its activity and the distribution law of AGPase phosphorylation in the process of starch synthesis with approaches of biochemistry, molecular biology, phosphopreteomics and enzyme kinetics in the high starch inbred line 08641 which is widely used in the southwestern region. These studies will provide the theoretical basis for targeting modification of AGPase enzyme and lay the foundation for further molecular improvement on inbred line 08-641.
