中文摘要: 良好的荧光药物传输载体可以动态可视化地传输药物、跟踪靶向给药,在荧光诊断与治疗一体化应用中起着关键的作用,对于药物设计、筛选具有重要的指导意义,已经引起人们越来越广泛关注。本项目将新型材料ESIPT高分子聚合物"点"作为药物载体和荧光共振能量转移(FRET)荧光供体,具有光动力治疗性质的方酸菁染料通过包埋和共价键与其相结合形成分子组装体系,实现疾病的近红外荧光诊断与光动力治疗一体化。通过分子间FRET的发生和抑制,研究光照下分子在病毒细菌细胞识别前与病毒细菌细胞凋亡后的荧光变化,以及调控单线态氧产生,可视化跟踪整个微观过程。本项目的研究将进一步拓宽ESIPT高分子聚合物"点"和方酸菁染料在生物领域的应用范围,为新的药物的设计、筛选以及临床应用提供新的理论指导和科学依据。

英文摘要: Good fluorescent drug delivery carrier can transmit drugs, track targeted drug delivery through dynamic visualization. It has being a growing concern due to it plays a key role in an integrated application of fluorescent diagnosis and treatment, and in guiding for drug design and screening. In this project, an integrated application of fluorescent diagnosis and treatment is realized through a formation of molecular assembly based on squaraine dyes for photodynamic therapy drugs embedding or covalently linking with fluorescent ESIPT polymer dot acting as a drug carrier and donor of fluorescence resonance energy transfer. The whole process can be visually track through the different fluorescence output derived from FRET occurring during the bacteria recognition and FRET inhibiting after the cell apoptosis upon light excitation, and the modulation of the singlet oxygen production. The project will further broaden the application of the ESIPT polymer dot and squaraine dyes in the biological field, and provide new theoretical guidance and scientific basis for new drug design, screening and clinical application.
