中文摘要: 通过区分标记单核(巨噬细胞)和多核(粒细胞)吞噬细胞的Tg(coro1a:eGFP;lyz:Dsred)和其强大的尾鳍再生能力,本课题主要关注il1b在斑马鱼幼鱼尾鳍切除后巨噬细胞和粒细胞的迁徙及随后的尾鳍再生中所发挥的具体作用和相关机理。我们初步在斑马鱼中克隆了il1b并检测了它在尾鳍受损后的表达图谱。在il1b knockdown时,粒细胞在伤口的富集受到明显抑制,但巨噬细胞的影响不明显。今后将重点关注:1) 创建il1b的突变体并确认表型的特异性;2) il1b 对吞噬细胞尤其是粒细胞向伤口迁徙的行为学影响的具体阶段;3)il1b对尾鳍再生的影响;4)构建Tg(hsp70:GAL4-stop-14xUAS-il1b-14xUAS-CFP)、Tg(lyz:loxP-stop-loxP-il1b-MYC) 等品系,结合基因芯片结果阐明il1b的作用机理。

英文摘要: Mononuclear (marophage) and polymorphonuclear (granulocyte) phagocytes migration and their roles in the regeneration after tissue disruption are very important questions that are still not well answered. Taking advantage of Tg(coro1a:eGFP;lyz:Dsred) reporter line that can distinguish granulocyte from macrophage at the same time and the powerful regeneration ability of zebrafish, the roles and mechanisms of il1b in granulocyte & macrophage migration and fin regeneration after amputation will be well dissected in this project. The preliminary study indicated that knockdown of il1b in the morphants could lead to the obvious decrease of neutrophils in the wound after fin amputation , but did not show significant influence on macrophages, suggesting distinct mechanisms under the migration of these two subtypes. Future study will mainly focus on subsequent issues: 1) Creating il1b mutant fish by TALEN and confirm the phenotype of il1b knockdown; 2) Analyzing the influence of il1b on the process of phagocytes especially granulocyte migration towards the wound; 3) Studying the role of il1b in the fin regeneration; 4) Establishing Tg(hsp70:GAL4-stop-14xUAS-il1b-14xUAS-CFP)、Tg(lyz:loxP-stop-loxP-il1b-MYC) and Tg(mpeg1:loxP-stop-loxP-il1b-MYC)transgenic lines; 5) Digging out the molecules downstream of il1b by microarray;
