中文摘要: 盐胁迫导致植物细胞积累大量的活性氧(ROS),对细胞造成严重损伤。植物细胞中含有完整ROS清除系统(抗氧化酶系统及抗氧化剂),对维持细胞内氧化还原平衡至关重要。本项目研究的甜菜M14品系是含有野生白花甜菜第9号染色体的单体附加系,前期研究显示该品系具有较强的耐盐特性,是一个特殊的甜菜种质资源。蛋白质组学研究结果表明,在200、400 mmol/L NaCl胁迫下,甜菜M14品系抗氧化酶系统中有6个蛋白质上调(GO、PrxR、Trx、APX、MDAR、GST),1个蛋白质下调(CAT)。本项目拟从甜菜M14品系中分离得到编码这些蛋白质的基因;对它们的序列特征,表达模式进行研究;进一步构建RNA干扰载体,获得阳性转基因植株,并进行耐盐特性研究,同时对7个基因之间的相互作用进行评价。本项目对于研究甜菜M14品系抗氧化酶系统响应盐胁迫应答过程及挖掘该品系优质基因资源具有重要意义。

英文摘要: Salt stress can lead to the production of large amounts of reactive oxygen species (ROS) in plant cells. They cause serious damage to the cells. Plant cells have evolved complicated ROS scavenging system (including antioxidant enzymes and antioxidant metabolites),which play a vital role in maintaining the redox balance in cells. The special sugar beet germplasm ,sugar beet apomictic monosomic additional M14 line, carrying an alien chromosome 9 of Beta corolliflora Zoss.,showed strong salt stress tolerance in our previous studies. The preliminary comparative proteomics studies had shown that under 200, 400 mmol/L NaCl stress, six proteins of the antioxidant enzymes system (GO, PrxR, Trx, APX, MDAR, GST) were increased in M14 line leaves, and one protein (CAT) was decreased. This project is aimed to obtain genes which enconde these proteins in the sugar beet M14 lines. Their sequences structures and expression patterns will be characterized. Further study on salt tolerance of transgenic plants obtained by transforming the RNAi expression vector and the interaction between the seven genes will be evaluated. This project will help us to understand the mechanism of antioxidant enzyme system response to salt stress in sugar beet M14 line, which also help investigating valuable genes from the special sugar beet.
