中文摘要: HNF1α是维护细胞代谢正常的重要转录蛋白,它的失活会引发包括糖尿病、肿瘤等在内的多种代谢类疾病。前期研究结果显示HNF1αSer249是其重要的功能位点,该位点翻译后修饰的异常显著影响HNF1α的生物学活性,我们推测该位点可能是ATM调控细胞代谢的作用靶。本项目拟从分子与细胞水平及动物水平对HNF1α的作用机制进行深入研究:1)HNF1αSer249突变体的基本特性(转录活性、DNA结合活性、定位)及与ATM的关系;2)HNF1αSer249突变对胰岛细胞系的糖代谢水平、ATP水平、Ca2+浓度、以及胰岛素水平等的影响;3)构建过表达HNF1αSer249突变体的转基因小鼠,检测其体重、血糖、胰岛素、葡萄糖代谢相关基因表达等指标4)ATM与HNF1A的相互作用。这为我们深入研究异常代谢并由其引发的重大疾病的分子机制,寻找新的、更有效的靶点蛋白,为早期诊断和防治这些重大疾病提供新的手段和

英文摘要: Hepatocyte nuclear factor-1 alpha (HNF1α) plays an important role in the regulation of a large number of genes expressed in the liver and pancreatic β-cells. However, the molecular mechanism involved in HNF1α-dependent gene activation have remained unclear. We now reported a phosphorylation sites in HNF1α: Ser249, which is involved in regulating HNF1α activation, especially in glucose-stimulated insulin secretion. We further identified that ATM, the protein product of the ATM gene that is mutated in the disease ataxia telangiectasia, phosphorylates HNF1α at Ser 249 in vitro and IR treatment induces phosphorylation of HNF1α at Ser 249 in vivo in an ATM-dependent manner. Taken together, these results support a model that phosphorylation processes are involved in the regulation of transactivity of HNF1α and provide new insights into understanding the role of HNF1α in maintaining glucose homeostasis
